Abstract Epilepsy is a neurological disease characterized by excessive and abnormal hyper-synchrony of electrical discharges the brain predisposition to generate epileptic seizures resulting in broad spectrum neurobiological insults, imposing psychological, cognitive, social also economic burdens sufferer. Voltage-gated sodium channels (VGSCs) are essential for generation propagation action potentials throughout central nervous system. Dysfunction these has been implicated pathogenesis epilepsy. VGSC inhibitors have demonstrated act as anticonvulsants suppress neuronal firing underlying seizures, used management treatment both genetic-idiopathic acquired epilepsies. We discuss forms idiopathic epilepsies caused mutations therapeutic efficacy blockers idiopathic, pharmacoresistant epilepsy this review. conclude that there need better alternative therapies can be alone or combination with The current anti-seizure medications (ASMs) especially some other types not yielded expected partly because they do show subtype-selectivity blocking while bringing side effects. Therefore, develop novel drug cocktails enhanced selectivity specific isoforms, achieve seizures.
